β乳糖蛋白抗體IgGELISA檢測(cè)試劑盒操作方法
雙抗體夾心法
1.包被：用0.05MPH9.牰碳酸鹽包被緩沖液將抗體稀釋蛋白質(zhì)含量為1～10μg/ml.在每個(gè)聚苯乙烯板的反應(yīng)孔中加0.1ml,4℃隔夜.次日,棄去孔內(nèi)溶液,用洗滌緩沖液洗3次,每次3分鐘.（簡(jiǎn)稱洗滌,下同）.
2. 加樣：加一定稀釋的待檢樣品0.1ml于上述已包被之反應(yīng)孔中,置37℃孵育1小時(shí).然后洗滌.（同時(shí)做空白孔,陰性對(duì)照孔及陽(yáng)性對(duì)照孔）.
3. 加酶標(biāo)抗體：于各反應(yīng)孔中,加入新鮮稀釋的酶標(biāo)抗體（經(jīng)滴定后的稀釋度）0.1ml.37℃孵育0.5～1小時(shí),洗滌.
4. 加底物液顯色：于各反應(yīng)孔中加入臨時(shí)配制的TMB底物溶液0.1ml,37℃10～30分鐘.
5. 終止反應(yīng)：于各反應(yīng)孔中加入2M硫酸0.05ml.
6. 結(jié)果判定：可于白色背景上,直接用肉眼觀察結(jié)果：反應(yīng)孔內(nèi)顏色越深,陽(yáng)性程度越強(qiáng),陰性反應(yīng)為無(wú)色或極淺,依據(jù)所呈顏色的深淺,以“+”、“-”號(hào)表示.也可測(cè)OD值：在ELISA檢測(cè)儀上,于450nm（若以ABTS顯色,則410nm）處,以空白對(duì)照孔調(diào)零后測(cè)各孔OD值,若大于規(guī)定的陰性對(duì)照OD值的2.1倍,即為陽(yáng)性.
間接法
1.用包被緩沖液將已知抗原稀釋1～10μg/ml,每孔加0.1ml,4℃隔夜；
2.次日洗滌3次；
3.加一定稀釋的待檢樣品（未知抗體）0.1ml于上述已包被之反應(yīng)孔中,置37℃孵育1小時(shí),洗滌；
4.（同時(shí)做空白、陰性及陽(yáng)性孔對(duì)照）于反應(yīng)孔中,加入新鮮稀釋的酶標(biāo)第二抗體（抗抗體）0.1ml；
5.37℃孵育35-60分鐘,洗滌；
6.’zui后一遍用DDW洗滌.
其余步驟同“雙抗體夾心法”的4、5、6.

應(yīng)用領(lǐng)域
ELISA法是免疫診斷中的一項(xiàng)新技術(shù),現(xiàn)已成功地應(yīng)用于多種病原微生物所引起的傳染病、寄生蟲(chóng)病及非傳染病等方面的免疫診斷.也已應(yīng)用于大分子抗原和小分子抗原的定量測(cè)定,根據(jù)已經(jīng)使用的結(jié)果,認(rèn)為ELISA法具有靈敏、特異、簡(jiǎn)單、快速、穩(wěn)定及易于自動(dòng)化操作等特點(diǎn).不僅適用于臨床標(biāo)本的檢查,而且由于一天之內(nèi)可以檢查幾百甚上千份標(biāo)本,因此,也適合于血清流行病學(xué)調(diào)查.本法不僅可以用來(lái)測(cè)定抗體,而且也可用于測(cè)定體液中的循環(huán)抗原,所以也是一種早期診斷的良好方法.因此ELISA法在生物醫(yī)學(xué)各領(lǐng)域的應(yīng)用范圍日益擴(kuò)大,
可概括四個(gè)方面：
1、免疫酶染色各種細(xì)胞內(nèi)成份的定位.
2、研究抗酶抗體的合成.
3、顯現(xiàn)微量的免疫沉淀反應(yīng).
4、定量檢測(cè)體液中抗原或抗體成份.
