Source

Recombinant human FGF acidic （rh-aFGF） Phe16-Asp155 （Accession # NP_000791） was produced in E.coli at ACRObiosystems.

Molecular Characterization

rh-aFGF contains no "Tags" and has a calculated MW of 15.8 kDa. DTT-reduced protein migrates as a 16 kDa polypeptide and the non-reduced migrates as a 16 kDa protein.

Purity

>98% as determined by SDS-PAGE. All lots are greater than 98% pure.

SDS-PAGE

Human aFGF
The purity of rh-aFGF was determined by SDS-PAGE,staining overnight with Coomassie Blue.

Bioactivity

The bioactivity of rh-aFGF protein was determined in a BALB/c 3T3 mouse fibroblasts cell proliferation assay. The ED50 of each lot is between 50-200 pg/ml.

Endotoxin

Less than 1.0 EU per 1 μg of the rh-aFGF by the LAL method.

Formulation

Bulk protein in a 0.22 μm filtered solution of PBS, pH 7.4 and delivered as liquid formulation or lyophilized powder. Normally 5-8% trehalose and mannitol are added as protectants before lyophilization.

Background

Heparin-binding growth factor 1 is a protein that in humans is encoded by the FGF1 gene. The protein encoded by this gene is a member of the fibroblast growth factor （FGF） family. FGF acidic is a potent growth factor for fibroblasts and endothelial cells （1）. FGF acidic is involved in wound repair, angiogenesis, and development （1）. FGF acidic is secreted from cells via an endoplasmic reticulum/Golgi independent mechanism （1,2）. The ability of FGF acidic to bind to heparin sulfate is required for its ability to interact with FGF receptors and induce signaling （1-4）. There are four distinct FGF receptors and each has multiple splice variants （1,3）. FGF acidic binds with high affinity to many, but not all, FGFRs （1）. Signaling cascades activated through FGF basic binding to FGFR include the ras-raf-MAPK, PLCγ/PKC, and PI3K/Akt pathways （1）.

Storage

Store at -20℃ in lyophilized state after receipt. For long term storage, upon reconstitution rh-aFGF should be aliquot and store at -20℃ or -80℃. Avoid repeated freeze-thaw cycles.
