一、基本描述：（CIS 誘導(dǎo)液）
CopyCutter Induction Solution是一種用于動態(tài)調(diào)控大腸桿菌質(zhì)?？截悢?shù)的分子生物學(xué)試劑。
其核心設(shè)計目的是解決不穩(wěn)定DNA序列或毒性基因的克隆難題，同時兼顧質(zhì)粒產(chǎn)量的提升。
EPI400菌株：通過改造宿主菌的基因?qū)崿F(xiàn)調(diào)控。
該菌株刪除了組成型表達(dá)的pcnB基因，替換為誘導(dǎo)型啟動子控制的改良pcnB基因。
未誘導(dǎo)時質(zhì)粒維持低拷貝，加入誘導(dǎo)液后激活表達(dá)，顯著提高質(zhì)粒產(chǎn)量。
EPI300菌株：誘導(dǎo)液激活突變的trfA基因，促進(jìn)含oriV復(fù)制子的質(zhì)粒高拷貝擴(kuò)增。
以無菌過濾的濃縮液形式提供（常見200X或1000X濃度），可直接加入培養(yǎng)基使用。
保存條件：-20℃長期穩(wěn)定（≥2年），4℃保存限6個月；若渾濁需用0.2-0.45μm濾膜過濾。
CopyCutter Induction Solution 使用優(yōu)點：
（1）CopyControl pCC1FOS和pCC2FOS裁體：線性化,去磷酸化,純化,已經(jīng)為連接做好準(zhǔn)備。
（2）不需要限制性內(nèi)切酶消化或脈沖凝膠電泳來準(zhǔn)備用于克隆的基因組DNA。
（3）使用pCC2FOS裁體可以較大化高通量末端測序結(jié)果。
（4）可以誘導(dǎo)單拷貝到每個細(xì)胞50拷貝。安全地獲得較高的DNA產(chǎn)量,同時保持了穩(wěn)定克隆單拷貝數(shù)。
（5）高效率的lambda包裝蛋白消除了背景值假陽性。
二、注意事項Notes:
（1）Competent cells must be transported on dry ice. Competent cells should be stored at -80C, and should not be repeatedly freeze-thawed and left for too long, otherwise the transformation efficiency of competent cells will be reduced.
（2）It is best to melt competent cells on ice.
（3）When performing transformation operations, it should be carried out at the corresponding temperature and sterile conditions.
（4）Transforming high concentrations of plasmids correspondingly reduces the amount of bacteria that end up being used for coating plates.
（5）To prevent unsuccessful conversion experiments, part of the junction reaction solution can be retained for reconversion to minimize losses.
（6）The product package contains the vector pUC19 DNA (10pg/ul) for control experiments.
