【簡單概述】
The DQ-BSA-Red lysosomal activity detection green fluorescent probe is a molecular probe based on bovine serum albumin (BSA) for fluorescence-labeled lysosomal activity.
High-concentration fluorescent probes are used to tag BSA molecules. Due to the high concentration of fluorescent probes, the labeled BSA molecules exhibit strong fluorescence self-inhibition, resulting in very low background fluorescence intensity.
In normal active lysosomes, DQ-BSA-Red degrades into BSA protein fragments, reducing the concentration of fluorescent molecules and thereby releasing the dye's self-inhibitory effect, enabling bright fluorescence emission in cells.
The functionally inactivated lysosomes were unable to degrade the BSA protein, and the self-inhibition between the high concentration of fluorescent molecules made the functionally inactivated lysosomes have no fluorescence.
The use of DQ-BSA-Red for lysosomal staining is more intuitive and can be directly imaged in living cells, eliminating the need for complex operations such as slide preparation, staining, cell lysis, centrifugation and detection of activity.
The dye is added to living cells 6 hours prior to staining, and imaging can be performed immediately after 6 hours. The DQ-BSA-Red dye is taken up into cells and exhibits extremely low background fluorescence before hydrolysis. Upon reaching lysosomes, it undergoes hydrolysis and emits a specific, bright fluorescence within lysosomes.
At the same time, the fluorescence spectrum peak width of the dye is small, which is compatible with other proteins or dyes of different wavelengths for co-staining; the dye has good photothermal stability and chemical stability, and can be stably imaged for a long time under confocal microscope, which is more stable than lysotrackor during imaging.
DQ-BSA-Red溶酶體活性檢測綠色熒光探針是基于牛血清白蛋白(BSA)的熒光溶酶體活性分子探針,使用高濃度的熒光探針對BSA進行標(biāo)記,標(biāo)記后的BSA分子由于高濃度的熒光探針存在高度的熒光自抑制現(xiàn)象, 因而本底熒光強度很低,DQ-BSA-Red在正常的活性溶酶體中降解會產(chǎn)生BSA蛋白質(zhì)片段,使熒光分子濃度變小從而解除染料的自抑制效果,在細胞中發(fā)出明亮的熒光。
功能失活的溶酶體則無法降解BSA蛋白,高濃度熒光分子間的自抑作用使得功能失活的溶酶體沒有熒光。
使用DQ-BSA-Red對溶酶體染色更加直觀,可直接進行活細胞成像,免除了制片、染色或者細胞裂解、離心檢測活性等復(fù)雜操作。
只需在檢測前6小時將染料加入活細胞中進行染色,6小時后直接成像即可。
DQ-BSA-Red染料可被內(nèi)吞進入細胞,水解前具有極低的熒光背景,到達溶酶體水解后能在溶酶體發(fā)出特異且明亮的熒光。
同時,染料的熒光光譜峰寬較小,兼容其他蛋白或者波長的染料進行共染色;染料具有較好的光熱穩(wěn)定性和化學(xué)穩(wěn)定性,在共聚焦顯微鏡下可穩(wěn)定成像較長時間,相較于lysotrackor在成像過程更加穩(wěn)定。
