【簡(jiǎn)單概述】（iFluor 488羊抗兔免疫球蛋白）

iFluor 488 Sheep anti-Rabbit Immunoglobulin (H+L) Fluorescently labeled secondary antibody. AAT Bioquest's iFluor dye is optimized for labeling proteins (particularly antibodies).

These dyes are bright, light stable and have little quenching effect on proteins.

They can be well excited by the main excitation light of the fluorescence instrument. The maximum fluorescence excitation and emission values of iFluor 488 goat anti-rabbit IgG (H + L) conjugate are approximately 491 nm and 514 nm, respectively.

These spectral characteristics make it an alternative to AlexaFluor488 goat anti-rabbit IgG (H + L) conjugate.

iFluor 488羊抗兔免疫球蛋白(H+L)熒光標(biāo)記二抗，AAT Bioquest的iFluor 染料針對(duì)標(biāo)記蛋白質(zhì)（特別是抗體）進(jìn)行了優(yōu)化。

這些染料是明亮的，光穩(wěn)定的，并且對(duì)蛋白質(zhì)的淬滅作用小。

它們可以被熒光儀器的主要激發(fā)光線很好地激發(fā)。iFluor 488山羊抗兔IgG（H + L）偶聯(lián)物的熒光激發(fā)和發(fā)射大值分別為?491 nm和?514 nm。

【操作步驟】

（一）孵育條件：

將稀釋后的二抗滴加到已完成一抗孵育的樣本（細(xì)胞或組織）上。

室溫孵育 30分鐘（避光操作）。

部分實(shí)驗(yàn)可調(diào)整至 4°C~37°C 孵育，但室溫是標(biāo)準(zhǔn)條件。

（二）清洗：

用 PBS 或 HBSS 緩沖液清洗樣本 2-3次，每次5-10分鐘，去除未結(jié)合抗體。

（三）復(fù)染（可選）：

若需細(xì)胞核染色，可加入DAPI（如100 nM）復(fù)染30秒，再用PBS清洗。

（四）封片與觀察：

用抗淬滅封片劑（如Fluoromount-G）封片，避光保存。

使用熒光顯微鏡觀察，選擇 Ex/Em=491/516 nm（iFluor 488）或標(biāo)準(zhǔn)FITC濾光片。
